ELISA (Enzyme-Linked Immunosorbent Assay) is a versatile testing technique. The ELISA immunoassays allow researchers to quantify a specific molecule of interest using antibodies. Typically, an enzyme (e.g., horseradish peroxidase) is either directly or indirectly coupled to the antibody to provide the detection signal. The choice of antibody and detection method will determine the sensitivity and specificity of the assay.

How our ELISAs are done.
With our ELISA service, we can quantify antigens or antibodies from a variety of virus-infected samples (e.g., cell lysate, tissue, and serum from human, animal, and plant specimens). We’re committed to providing simple, robust, and cost-effective ELISA services to the research community. At VRS, we provide multiple advanced ELISA services, including:
Direct ELISA: In direct ELISA, the antigen is immobilized to a multi-well plate and is detected by a specific conjugated primary antibody.
Indirect ELISA: In indirect ELISA, the antigen is immobilized to a multi-well plate and is detected first by a specific unconjugated primary antibody. Next, an enzyme-conjugated secondary antibody is applied, directed against the host species of the primary antibody.
Sandwich ELISA: In sandwich ELISA, a capture antibody is immobilized to a multi-well plate. The sample is then added, followed by a detection antibody. The capture and detection antibodies are specific for different and non-overlapping epitopes of the antigen. Both direct and indirect methods can be used when performing sandwich ELISA. Sandwich ELISA is used for accurate detection of antigen concentration in an unknown sample and when high detection sensitivity and specificity are required.
Competition ELISA (also called inhibition ELISA): In competition ELISA, an antigen is immobilized to a multi-well plate and incubated with an antibody that has been previously incubated with the sample-of-interest (that should also contain that specific antigen).
The sample antigen competes with a reference antigen for binding to a limited amount of antibody. The concentration of the antigen in a sample inversely correlates with the signal output: the higher the sample antigen concentration, the weaker the eventual signal. Both direct and indirect methods can be used.
Competitive ELISA is used to measure the concentration of a small molecule in a crude or complex sample when only one antibody is available for the antigen of interest.
