Determine the Antiviral Potency of Your Compound

The 50% inhibitory concentration (IC50) is used to quantify the potency of an antiviral compound by determining the concentration required to reduce viral infection by 50%.

Compounds are tested across a range of concentrations in virus-infected cells, allowing a dose-response curve to be generated. Determining the IC50 provides a quantitative measure of antiviral activity and can be used to compare the potency of different compounds or formulations.

Our Approach

To determine the IC50, susceptible host cells are infected with the virus and treated with a range of concentrations of the test compound under defined experimental conditions.

At the end of the infection period, cells are fixed and immunostained for a viral antigen. Viral infection is quantified using high-content imaging, allowing the percentage of infected cells at each compound concentration to be determined.

The antiviral activity at each concentration is compared with the appropriate infected and uninfected controls to generate a dose-response curve, from which the 50% inhibitory concentration (IC50) is calculated.

Cytotoxicity is assessed in parallel to distinguish genuine antiviral activity from reductions in infection caused by compound toxicity and to determine the selectivity of the test compound.

Different viruses, cell lines, compound concentrations and treatment conditions can be evaluated according to the objectives of the study.
